    the probe is designed to only bind to certain sequences, for example it can be designed to only bind to a DNA sequence with a certain missense mutation and activate the fluorescence

    2022, February 1st, look at the mess out country has become due to this fentynl. Heroin was one thing. It was actually pretty darn safe. It's long gone. Here in Detroit it's been gone for 5 years, only fentynl since then. It has no euphoria, none of the feel good of heroin, just a nasty drug that either puts you to sleep or if it don't you really don't feel it..... or the 3rd option- you die. It seems to be not only 50x stronger than heroin.....but 50x harder to quit. Horrid. 21 of my 40 years stuck on IV heroin and fentynl. My brain just doesn't work without it anymore.

      At the very least with methadone, you always have a wake up, rarely go sick, and you can eventually work up to having to go only once a month. Street dope, has to many negative longterm outcomes. Mitigate those issues & you can have a more sane healthy life.

    I wonder if the probe-oligonucleotide will be denatured (separated from quencher) during the first round of PCR and emit signal ? or the probe has to bind on the template DNA to emit signal ?

      From my perspective, the probe, can emit signal in the first place, but it is inhibited by quencher. Therefore, in the first round the X/Y complement haven't been created yet. The probe will eventually end up with the quencher, but in the third round, the complement is created and the probe bind to it, in the other words, the probe is permanently separated from sequence which allow it to emit the signal.

      I think the signal is only read at the end of the PCR-reaction. At this stage, when the temperature is lowered, the unbound reporters will bind again with the quenchers. Therefore no signal is emitted when the results are interpretted.

      In order to begin to identify the effect of a given SNP in a population, you would need to at least have the disease susceptibility data, and the genotypic data for the same individuals, and then run a correlation between the susceptibility and the given SNP states. Look up 'genome wide association studies' (GWAS), and 'Quantitative Trait Loci' (QTL) mapping studies in order to see some of the common methods used for this. Bear in mind, the effect is the difference between the average with 1 allele and the average with the other allele. A SNP is only relevant for diagnosing susceptibility in a population if the effect is both statistically significant, and quantitatively sizable. Granted, if the SNP itself is causative for a specific phenotype, you'd want to study the effect with as little background noise as possible, and looking at back-crossed mutants when compared to wild-type individuals might then be optimal.
